  • UPLC測定黃連中鹽酸小檗堿和鹽酸巴馬汀的含量

    摘  要:建立UPLC測定黃連中鹽酸小檗堿和鹽酸巴馬汀含量的方法。采用ACQUITY UPLC BEH-C18色譜柱(2.1 mm×50 mm×1.7 μm),以乙腈-0.05 mol/L磷酸二氫鉀溶液為流動相,梯度洗脫,流速為0.2 mL/min,檢測波長為350 nm。結果表明鹽酸小檗堿和鹽酸巴馬汀分別在0.037 2~0.297 6 μg/mL,0.008 2~0.065 6 μg/mL的濃度范圍內(nèi)均有良好的線性關系(r=0.999 9),回收率均大于95%,RSD均小于2%。該方法快速、準確、重現(xiàn)性好,可用于測定黃連中鹽酸小檗堿和鹽酸巴馬汀的含量。

    UPLC simultaneous determination of berberine hydrochloride and
    palmatine hydrochloride in Coptis

    Abstract: A method was established for simultaneously determining berberine hydrochloride and palmatine hydrochloride in Coptis by UPLC. The optimal conditions of separation and detection were achieved on an ACQUITY UPLC BEH-C18 column (2.1 mm×50 mm×1.7 μm) eluted with a gradient acetonitrile and 0.05 mol/L potassium dihydrogen phosphate solution at a flow rate of 0.2 mL/min, and detected at 350 nm. The results showed that there were good linear relationships (r=0.999 9) when the concentration of berberine hydrochloride and palmatine hydrochloride were in the range of 0.0372-0.2976 μg/mL and 0.0082-0.0656 μg/mL respectively, the recovery rates were more than 95%, and RSDs were less than 2%. This method was simple, rapid, accurate, and reproducible, and it can be used for determining the concentration of berberine hydrochloride and palmatine hydrochloride in Coptis.
